To get the TMP weapon in Resident Evil 4 Remake, remove all your items before entering the Village Square Gate. Remember, the location of the TMP gun will not be available for you even if you leave a single gun weapon or items in your Inventory.
You will find a well open up as soon as you have removed all your items. The well will be located on the south side of the Village behind the barn.
You will find a ladder on the right side of the cart that you need to drop down from. Remember that the coach will be on the path of the Village Entrance itself. Once inside the tunnel, you will find a chest. Open up the chest and find the Level 3 Upgraded TMP. Return to the ground near the tower again using the other entrance.

The best way to use the TMP is close Range firing, primarily because of TMP’s low Precision, but it can hit accurately at lower distances. But that being said, dont go too near to your enemies, as they will grab you if they somehow dodge your bullets.
